For influenza viruses, the two glycoproteins hemagglutinin (HA) and neuraminidase (NA) play important roles in receptor binding, membrane fusion and the release of progeny virions. Due to a significant genetic separation (including length variation) among subtypes, a universal numbering scheme is useful in assessing the amino acid equivalency for various subtypes of interest. In the year 2014, Burke and Smith analyzed the structural alignment among 18 HA sequences by including a number of HA PDB records for recommending an HA numbering scheme. Afterward, several HA subtype PDB records became newly available, and the HA alignment of influenza A virus was not updated based on our knowledge. Thus, a comprehensive analysis together with entire data is required to update the HA numbering scheme, as well as to establish an NA numbering scheme that was yet proposed. In this work, the updated (till 2019) 18 HA- and 11 NA-sequence alignments are investigated based on mature proteins. The reporting updated HA alignment was found similarly matching with Burke and Smith work. Therefore, the similar HA sequence alignment procedure is followed to compare 11 NA subtypes. The known structures of entire NA subtypes enable us to assess amino acids sequences which are functionally and structurally comparable. Based on our knowledge, this is the preliminary report to implement 11 NA subtypes sequence alignment (up to 2019). Thereby, the improved understanding of the biological functions of both HA and NA subtypes in the influenza virus, in terms of amino acid equivalency, could facilitate a marginal design for antivirals to struggle influenza virus infections.
